A versatile approach for cell health determination and compound profiling

Whether screening for cytotoxic or cytoprotective agents, or to identify potential off-target drug effects early, multiplexed cell health assays play a key role within drug discovery. The extent of cell death can be determined in simple live/dead screens using standard dyes (such as calcein-AM and propidium iodide).

Deeper profiling to distinguish between the various pathways may be achieved through further multiplexing (caspase, annexin V, nuclear condensation, mitochondrial health) or multiparametric readouts (cell proliferation and cell cycle) to provide valuable insights into mechanism of action.

The effect of compounds on the cell cycle is a widely studied application. Traditionally, flow cytometry is used to characterise DNA content, however, plate-based imaging using acumen Cellista® is more amenable to automation.

DNA binding dyes used to quantify cell cycle phases may be re-analysed over time to monitor kinetics, or multiplexed to identify the expression of disease relevant proteins or cell cycle markers. Adherent cells are analysed in-situ to preserve their morphology and results normalised to total cell number highlight cytotoxicity or cytostasis.

Signalling cascades have traditionally been monitored by biochemical and lysate-based assays, however these approaches offer low content data reporting.

Speeding up angiogenesis detection assays

Angiogenesis is a fundamental multistep process by which new capillary blood vessels grow from pre-existing vasculature and is critical in the progression of tumours moving from dormancy to malignancy.

Using a microscope-based approach, angiogenic tube formation assays require entire well areas to be analysed in order to gain robust, statistically significant data, however, this can result in lengthy microplate read times. We have developed a simple application for analysing tube formation from the entire well area of microplates in just five minutes. Open source TIFF images from wells of interest may be exported for in depth analysis using third party image analysis packages.
