Acumen Applications - RNAi Profiling

The recent discovery that small interfering RNA (siRNA) technology has potent and rapid gene silencing properties has led to its rapid adoption by the scientific community for the identification of candidate drug target genes. MicroRNAs (miRNAs) are a class of small RNAs that can modulate mRNA expression in mammalian cells by blocking mRNA translation.

In collaboration with Ambion, TTP LabTech have profiled the phenotype of both siRNA and miRNA libraries in HeLa cells using cell proliferation in which the total cell number was determined using propidium iodide staining. Analysis of samples was rapid, taking about 10 minutes to process a 96 well microplate.

The features of the Acumen eX3 are highly compatible for the phenotypic profiling of RNAi libraries. These include determination of the effects of RNA inhibitors on cell proliferation, essential for data normalisation.
